HPLC法测定N-羟基-N-(2-((1-(4-氯苯基)-1H-吡唑-3-基)氧)甲基)苯基氨基甲酸甲酯

摘 要:采用Aglient Extend-C18 5 μm 250 mm×4.6 mm色谱柱,流动相为甲醇-0.4%磷酸溶液(v∶v=75∶25),流速为1 mL/min,275 nm为检测波长,建立了测定N-羟基-N-(2-((1-(4-氯苯基)-1H-吡唑-3-基)氧)甲基)苯基氨基甲酸甲酯含量的高效液相色谱分析方法。在进样量为0.53~3.52 μg的范围内线性关系良好(y=125679.4x+15010.4,r=0.9999),RSD 0.24%,回收率99.2%~100.4%,分析周期为15 min。方法快速、简便、准确,可用于N-羟基-N-(2-((1-(4-氯苯基)-1H-吡唑-3-基)氧)甲基)苯基氨基甲酸甲酯反应液及产品的质量控制

Determination of methyl 2-((1-(4-chlorophenyl)-1H-pyrazol-3-yloxy)methyl)phenyl(hydroxy)carbamate by HPLC

Abstract:A HPLC method was established for the determination of methyl 2-((1-(4-chlorophenyl)-1H-pyrazol-3-yloxy)methyl)phenyl(hydroxy)carbamate. The HPLC system consisted of Aglient Exlend-C18 column(5 μm, 4.6 mm×150 mm), a mixture of methanol and 0.4% phosphoric acid aquas solution at a volume-ratio of 75 to 25 as mobile phase, 275 nm as the detected wavelength, a flow rate of 1mL/min and column temperature of 40℃. The experimental results showed that this method for the determination of methyl 2-((1-(4-chlorophenyl)-1H-pyrazol-3-yloxy)methyl)phenyl(hydroxy)carbamate had a good linear correlation in a rang from 0. 53 μg to 3.52 μg(y=1 256 794.4x+15 010.4; r=0.999 9). RSD was 0.24% and the recovery was in a rang from 99.2% to 100.4%. The complete period of analysis was 15 min. This method was proved to be sensitive, fast, and accurate. It could be applied in the quality control of methyl 2-((1-(4-chlorophenyl)-1H-pyrazol-3-yloxy)methyl)phenyl(hydroxy)carbamate.
