阿托伐他汀钙的合成工艺改进

摘 要:以(4R-cis)-6-[2-[2-(4-氟苯基)-5-(1-异丙基)-3-苯基-4-[(苯胺)-羰基-1H-吡咯-1-基]乙基]-2,2-二甲基-1,3-二氧戊环-4-乙酸叔丁酯为起始原料,通过酸水解、碱水解,与醋酸钙中和3步得到目标化合物阿托伐他汀钙,研究了酸、溶剂和温度对反应的影响规律。优化反应条件为:水解采用盐酸,反应介质为乙醇,反应温度为 55℃。在上述条件下,产品收率达88.7%。产物结构经核磁共振氢谱(1H NMR)和质谱(ESI-MS)确证。

An Improved Process for the Synthesis of Atorvastain Calium

Abstract:The target compound was synthesized from (4R-cis)-1,1-dimethylethyl-6-[2[2-4(fluorophenyl)-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)-carbonyl]-H-pyrrollyl]ethyl]-2,2-dimethyl-1,3-dioxane-4-acetate via three steps including acid hydrolysis, alkali hydrolysis and calcium acetate neutralization. The effects of acid, reaction medium and temperature on the reaction were studied. The results showed that the optimum reaction condition are as follows: hydrochloric acid as the hydrolyzation acid, ethanol as the solvent of reaction, the reaction temperature of 55℃. Under above conditions, the total yield was 88.7%. The structure of atorvastain calium was verified by 1H NMR and ESI-MS.
