苯乙烯吡唑类化合物的合成及体外抗增殖活性

摘 要:以3,4-二甲氧基苯甲醛为原料,经羟醛缩合、环合、烷基化、亲核取代等反应合成5个未见文献报道的苯乙烯吡唑类化合物,结构均经核磁共振氢谱和质谱确证,以MTT法考察目标化合物对4种肿瘤细胞的增殖抑制作用。其中化合物5b~5e对肿瘤细胞HT29的增殖抑制作用强于先导化合物姜黄素。化合物5a和5d抑制A375细胞增殖的IC50值与姜黄素相当。

Synthesis and Anti-proliferative Activity of Styryl Pyrazole Derivatives

Abstract:Five styryl pyrazole derivatives were synthesized via aldol condensation, cyclization, alkylation and nucleophilic substitution reactions using 3,4-dimethoxybenzaldehyde as the starting material. The structures of the obtained styryl pyrazole derivatives were confirmed by 1H NMR and MS spectra. The growth inhibitory effects of these compounds were evaluated by MTT assay in four kinds of cell lines. Compounds 5b~5e showed improved anti-proliferative effects in HT29 cell line compared to that of curcumin while compound 5a and 5d showed similar potency against A375 cell line as curcumin.
