丹参-葛根提取物抗氧化能力与活性成分的相关性研究 |

摘 要:为揭示丹参-葛根提取物抗氧化作用的活性成分,以DPPH自由基清除率和总还原能力(FRAP)法评价提取物的抗氧化活性,建立UPLC分离方法鉴别其成分,采用Minitab相关性分析和偏最小二乘回归法(PLS)分析提取物抗氧化能力与成分的相关性。结果表明,在提取物抗氧化性与单一成分相关性的研究中,大豆苷对抗氧化性的贡献低。葛根素、丹酚酸B、迷迭香酸与DPPH清除率的相关性较强,相关系数R约为0.6,高于FRAP值的相关性(R约为0.5),说明其主要通过清除自由基达到抗氧化作用。以葛根素、丹酚酸B、迷迭香酸、大豆苷4种组分为变量与DPPH清除率建立的关系模型中,R接近0.7,呈明显的正相关,表明这四种组分是丹参-葛根提取物抗氧化活性的主要贡献者。 |

Study on the Correlation of Antioxidant Activity with Active Components Extracted from Salviae Miltiorrhizae and Radix Puerariae |

Abstract:DPPH free radical-scavenging and Ferric reducing antioxidant power (FRAP) assays were chosen to evaluate the antioxidant activities of salviae miltiorrhizae and radix puerariae extracts. The UHPLC method was established for the analysis of active components in salvia miltiorrhiza and radix puerariae. Minitab correlation analysis and partial least squares (PLS) were proposed for analysis of the correlation between antioxidant activities and component contents. Results showed that daidzein had low contribution to antioxidant capacity of extracts. The correlations of puerarin,salvianolic acid B, rosmarinic acid and DPPH free radical-scavenging was better. The coefficients of correlation(R) was 0.6, which was higher than the correlation coefficient(R=0.5) of Ferric reducing antioxidant power (FRAP) and activate components. It showed that the antioxidation effect was mainly achieved by scavenging free radicals. The model took four components daidzein, puerarin, salvianolic acid B and rosmarinic acid as independent variables, DPPH free radical-scavenging as variables was established. The correlation coefficient of the model approached 0.7, which presented significant positive correlation. It showed these four components are the main contributors for the antioxidant activity of extract from salvia miltiorrhiza and radix pueraria. The results established that scientific foundation for the development and application of antioxidants. |
