摘　要：以3-异丁基戊二酸为起始原料，经脱水、氨解、霍夫曼重排、手性拆分、游离、重结晶，以38.1%总收率得普瑞巴林。工艺稳定可控，产品纯度高（＞99.9%），适合于工业化放大生产。同时，对产品进行了结构确证。

Preparation and Structure Confirmation of Pregabalin

Abstract：Pregabalin was synthesized from 3-isobutylglutaric acid. This process included dehydration, ammonolysis, Hofmann rearrangement, chiral separation, and recrystallization(total yield 38.1%). This improved process was stable and controllable, with high product purity (＞99.9%) and suitable for industrialization. Also, the structure of pregabalin was systematically identified.
