嘧啶高效降解菌MD2的筛选、鉴定及其条件优化

摘 要:从实验室嘧啶生化好氧池废水及其活性污泥中分离得到1株耐盐高效嘧啶降解菌MD2,该菌株可以嘧啶为唯一碳源生长。通过形态特征、生理生化反应、16S rDNA鉴定及序列比分析,确定菌株MD2为1株柠檬酸杆菌;正交实验确定MD2降解嘧啶的最佳环境条件组合为嘧啶浓度150 mg·L-1、盐度1%、pH值7.0、温度30℃,该条件下菌株MD2对嘧啶的降解率达到67.84%,比原来提高2.5%左右。

Isolation, Identification and Optimization of Pyrimidine-degrading Strain

Abstract:One salt-tolerant and high-efficiency pyrimidine-degrading strain was isolated from the wastewater and activated sludge in the laboratory biochemical aerobic tank, which was used for pyrimidine wastewater treatment. The strain could feed pyrimidine as the sole carbon source and named MD2. According to morphological characteristics, physiological and biochemical reactions, 16S rDNA identification and sequence ratio analysis, strain MD2 was identified as a strain of Citrobacter sp.. The optimum environmental conditions for pyrimidine degradation by MD2 was obtained by orthogonal experiments. The pyrimidine concentration was 100 mg·L-1, the salinity was 1%, the pH was 7.0, and the temperature was 30℃. Under the above condition, the degradation rate of pyrimidine by strain MD2 reached 67.84%, about 2.5% higher than before.
