均匀设计法优化栀子苷类活性成分提取工艺及其应用

摘 要:以栀子苷的溶出率为指标,采用均匀设计U10×(108)考察了回流和超声法中各提取条件对栀子苷类活性成分溶出的影响,进行回归分析建立数学模型,得到优化条件。结果表明超声法明显优于回流法,确定了以70%乙醇按照20 mL·g-1液固比超声提取50 min的提取条件。将该提取工艺用于10批栀子和10批焦栀子药材的提取,结果发现,栀子药材中栀子苷含量4.26%~6.29%。各批次焦栀子中栀子苷均低于栀子,含量在3.45%~5.23%。由此可见,采用均匀设计法可快速、准确地确定栀子苷类活性成分的最佳提取工艺、条件,能更准确地评价和控制栀子药材质量。

Optimization and Application of Extraction Process of Iridoids Components from Gardeniae Fructus by Uniform Design

Abstract:In this paper, uniform design U10*(108) was used to investigate the factors affecting the release of iridoids components from Gardeniae Fructus in both reflux and ultrasonic extraction methods using the amount of gardenoside as evaluation criteria. The extraction conditions including time, solvent (ethanol) concentration and liquid-to-solid ratio were considered. Regression analysis were performed to establish mathematical models. The results showed that ultrasonic extraction method was obviously superior to the reflux extraction method. The optimal conditions for ultrasonic extraction process were obtained as following: 70% ethanol was used as solvent; the liquid-solid ratio was of 20 mL·g-1; and the extracting time was of 50 min. The extraction conditions were tested in extraction of 10 batches of Gardenia and 10 batches of coke Gardenia samples. The results showed that the iridoid glycosides constituents were different in different batches of gardenia medicinal materials. The contents of gardenoside varied from 4.26% to 6.29%. The contents of gardenoside in different batches of coke gardenia were lower than those in gardenia with a range of 3.45% to 5.23%. Thus, uniform design was successfully used to appraise the appropriate method and conditions for quick and accurate extraction of iridoid components. This presented a basis for quality evaluation and precise control of gardenia.
