Fe3O4-CNTs复合物的制备及载药效果

摘 要:为充分利用Fe3O4和碳纳米管(CNTs)在靶向和载药方面的性能优势,采用化学原位沉积法制备了Fe3O4-CNTs磁靶向复合物,并通过物理法分别负载了阿司匹林(ASA)、庆大霉素(GM)和阿霉素(DOX)。通过CNTs上Fe3O4纳米粒子沉积情况来优化铁盐与CNTs不同质量比,通过单因素实验考察负载时间、负载温度、原料质量比对Fe3O4-CNTs载药量的影响。实验结果表明,当铁盐与CNTs质量比为4.5∶7.0时,Fe3O4颗粒均匀生长在CNTs表面,Fe3O4-CNTs复合物具有较好磁性;Fe3O4-CNTs复合物负载ASA时,在CNTs与ASA的质量比为1∶4、负载时间为120 min、负载温度为40 ℃条件下,最高载药量为4.85%;Fe3O4-CNTs复合物负载GM时,在CNTs与GM质量比为1∶4、负载时间为150 min、负载温度为30 ℃条件下,最高载药量为45.04%;Fe3O4-CNTs复合物负载DOX时,在CNTs与DOX的质量比为1.0∶1.5、负载时间为180 min、负载温度为30 ℃条件下,最高载药量为17.02%。

Preparation and Drug Loading of Fe3O4-CNTs Complex

Abstract:In order to make full use of the performance advantages of Fe3O4 and CNTs in targeting and loading prescriptions, Fe3O4-CNTs magnetic targeting complex was prepared by chemical in-situ deposition method, and ASA, GM and DOX were loaded on Fe3O4-CNTsby physical method, respectively. The different mass ratios of iron salt and CNTs were optimized by the deposition state of Fe3O4 nanoparticles on CNTs. The effects of loading time, loading temperature and material quality ratio on the loading of Fe3O4-CNTs were investigated by single factor experiment.The experimental results showed that when the mass of iron salt and CNTs was 4.5∶7.0, Fe3O4 particles grew uniformly on the surface of CNTs, and the Fe3O4-CNTs complex had good magnetic properties.When the Fe3O4-CNTs complex was loaded with ASA, the mass ratio of CNTs to ASA was 1∶4, the loading time was 120 min, and the loading temperature was 40 ℃, the maximum loading was 4.85%; When the Fe3O4-CNTs complex was loaded with GM, the mass ratio of CNTs and GM was 1:4, the loading time was 150 min, and the loading temperature was 30 ℃, the maximum loading was 45.04%; When the Fe3O4-CNTs complex was loaded with DOX, the ratio of CNTs to DOX was 1.0∶1.5, the loading time was 180 min, and the loading temperature was 30 ℃, the maximum loading was 17.02%.
