一种阿伐那非的杂质合成和表征

摘 要:在乙腈溶剂中以4-(3-氯-4-甲氧基苄基)氨基-2-甲硫基嘧啶-5-羧酸乙酯(2)为起始原料,加入Oxone氧化反应得到水解产物4-(3-氯-4-甲氧基苄基氨基)-2-羟基嘧啶-5-羧酸乙酯(7),4-(3-氯-4-甲氧基苄基氨基)-2-羟基嘧啶-5-羧酸乙酯(7)经碱液水解,调酸得到4-(3-氯-4-甲氧基苄基氨基)-2-羟基嘧啶-5-羧酸(8)。4-(3-氯-4-甲氧基苄基氨基)-2-羟基嘧啶-5-羧酸(8)与氯化亚砜反应生成酰氯后与2-氨甲基嘧啶甲磺酸盐反应得到杂质4-(3-氯-4-甲氧基苄基氨基)-2-羟基-N-((嘧啶-2-基)甲基)嘧啶-5-甲酰胺(1),总收率7.66%,并对此合成杂质进行了结构表征。

Synthesis and Characterization of an Impurity of Avanafil

Abstract:Ethyl 4-(3-chloro-4-methoxybenzyl) amino-2-(methylthio) pyrimidine-5-carboxylate (7) was prepared from 4-(3-chloro-4-methoxybenzylamino)-2-hydroxypyrimidine-5-carboxylate(2) by oxone oxidation reaction in acetonitrile. 4-(3-chloro-4-methoxybenzylamino)-2-hydroxypyrimidine-5-carboxylic acid(8) was obtained by hydrolysis of ethyl 4-(3-chloro-4-methoxybenzylamino)-2-hydroxypyrimidine-5-carboxyate (7) by alkali hydrolysis. 4-(3-chloro-4-methoxybenzylamino)-2-hydroxypyrimidine-5-carboxylic acid(8) reacted with thionyl chloride to form acyl chloride and reacted with 2-aminomethyl pyrimidine methanesulfonate to give 4-(3-chloro-4-methoxybenzylamino)-2-hydroxy-N-((pyrimidine-2-yl)methyl) pyrimidine-5-formamide(1) with a total yield of 7.66%. The structure of the synthetic impurity was characterized.
