5-羟基异喹啉-7-羧酸及其异构体7-羟基异喹啉-5-羧酸的合成 |

摘 要:对重要的医药中间体5-羟基异喹啉-7-羧酸和7-羟基异喹啉-5-羧酸的合成工艺进行了初步研究,以3,5-二溴苯甲醛为原料经过Pomeranz-Fritsch、甲基化、钯催化插羰、水解和脱甲基等多步反应得到目标化合物,产品的纯度分别为98.2%、97.8%,收率分别为86.3%、83.7%。其结构经1H NMR,13C NMR和LC-MS确证。 |

Synthesis of 5-Hydroxy Isoquinoline-7-carboxylic Acid and Its Isomer |

Abstract:The purpose of this paper is to study the synthetic process of the important pharmaceutical intermediates of 5-hydroxyisoquinoline-7-carboxylic acid and 7-hydroxyisoquinoline-5-carboxylic. The target compound and their isomers were prepared by 3,5-dibromobenzaldehyde via Pomeranz Fritsch synthesis, methylation, palladium catalyzed carbonylation, hydrolysis and demethylation. The purity of the product is 98.2%,97.8% and the yield is 86.3%, 83.7%. The structures of the target compounds were confirmed by 1H NMR, 13C NMR and LC-MS. |
