4-氯-2-氨基联苯的绿色合成与表征

摘 要:以对氯溴苯和邻氯硝基苯为原料,经格氏反应、Suzuki偶联、Pd-C催化加氢还原3步反应合成了4'-氯-2-氨基联苯。格氏反应中,优化的反应条件为n(对氯溴苯)∶n(硼酸三甲酯)=1.0∶1.2,反应温度为0 ℃。Suzuki偶联反应中,优化的反应条件为n(邻氯硝基苯)∶n(4-氯苯硼酸)∶n(PdCl2(PPh3)2)=1.00∶1.10:0.01,还原反应的较佳反应温度为50 ℃。在优化的反应条件下,4'-氯-2-氨基联苯的合成总收率≥59.7%(以对氯溴苯计),纯度95.0%;产物结构经IR、1H NMR、MS确证。该工艺简单经济,条件温和,高效环保,适合工业化生产。

Green Synthesis and Characterization of 4'-Chloro-2-aminobiphenyl

Abstract:4'-Chloro-2-aminobiphenyl was synthesized using 4-bromochlorobenzene and 1-chloro-2-nitrobenzene as starting material by a series of Grignard reaction, Suzuki coupling reaction, Pd-C catalytic hydrogenation reduction. Under the optimal reaction conditions, the total yield of 4'-chloro-2-aminobiphenyl was over 59.7% based on 4-bromochlorobenzen, the purity was 95.0%. The structure was confirmed by IR, 1H NMR, MS. In the first step, the ratio of 4-bromochlorobenzen∶trimethyl borate=1.0∶1.2(mol ratio) was the optimized condition, The optimal reaction temperature is 0 ℃. In the second step, the ratio of n(1-chloro-2-nitrobenzene)∶n(4-chlorophenylboronic acid)∶n(PdCl2(PPh3)2)=1.00∶1.10∶0.01 was the optimized condition. The optimal reaction temperature of the reduction reaction is 50 ℃. This process is simple, warm economical, efficient and environmental friendly, which is suitable for industrial scale manufacture.
