Cobicistat 关键中间体的合成工艺研究 |

摘 要:以 2-甲基硫代丙酰胺为原料, 经过 3 步反应得到 Cobicistat 关键中间体(S)-1-(2-异丙基噬哇-4-
基)甲基-1-甲基-3-(2-氧代四氢吠喃-3-基)腺, 反应总收率为 52.7 %[以 2-甲基硫代丙酰胺(2)计]。 中间体及产物结构经 1H NMR、 ESI-MS 表征。
确定了合成中间体 4 的适宜物料比为: n(甲胺盐酸盐):n(3)=2.0:
1.0。 优化了产物 1 的工艺条件: 物料配比为 n(CDI):n(5):n(4)=1.5:1.2:1.0, 反应溶剂为二氯甲炕, 反应时间 10 h。 优化条件下, 产物 1
的收率达 80.8 %。

Synthesis of Key Intermediate of Cobicistat

Abstract:(S)-1-(2-Isopropylthiazole-4-yl) methyl-1-methyl-3-(2-oxo-oxolane-3-yl) urea, the key
intermediate
of Cobicistat, was synthesized from 2-methylthio- propionamide in a three-step reaction, the total
yield was 52.7% (base on compound 2). The structures of intermediates and products were
characterized by 1H NMR and ESI-MS. The suitable material ratio of the synthetic intermediate 4 is
as followings: n (methylamine hydrochloride):n (3)=
2.0 :1.0; The mole ratio of the synthetic product 1 was of n (CDI):n (5) :n (4)=1.5 :1.2 :1.0; the
reaction solvent was
dichloromethane, the reaction time was 10 h, and the yield of product 1 was up to 80.8% under these
reaction conditions.
