盐酸普萘洛尔的合成

摘 要:以3-(1-萘氧基)-1,2-环氧丙烷和异丙胺为原料,合成了中间体普萘洛尔,经氯化氢酸化得到盐酸普萘洛尔。考察了催化剂种类、反应物配比、反应温度、反应时间对中间体普萘洛尔收率的影响。优化工艺条件为:n(3-(1-萘氧基)-1,2-环氧丙烷)∶n(异丙胺)=1.0∶1.2,催化剂为水,加入量为2 mL,反应温度为50 ℃,反应时间为4 h。优化条件下盐酸普萘洛尔收率为83.2%。

Synthesis of Propranolol Hydrochloride

Abstract:Using 3-(1-naphthyloxy)-1,2-epoxypropane and isopropylamine as raw materials, the intermediate propranolol was synthesized and acidified with hydrogen chloride to obtain propranolol hydrochloride. The effects of catalyst type, raw material ratio, reaction temperature and reaction time on the yield of the intermediate propranolol were investigated. The optimized process conditions are the followings: n[3-(1-naphthyloxy)-1,2-epoxypropane)]∶n(isopropylamine)=1.0∶1.2; the catalyst is water while the addition amount is 2 mL; the reaction temperature is 50 ℃; the reaction time is 4 h. Under optimal conditions, the yield of propranolol hydrochloride is 83.2%.
