(N,N-二甲基氨基)磷酰氯吗啉代核苷单体的合成工艺

摘 要:分别以胞苷、5-甲基尿苷为原料,经氨基保护、5'-OH保护、吗啉环形成、吗啉环N-三苯甲基化、羟基脱保护、羟基磷酰化6步反应合成得到2个磷酰胺吗啉代寡核苷酸单体—(N,N-二甲基氨基)磷酰氯吗啉代核苷系列物,反应总收率分别为20%和30%。中间体及目标产物经LC-MS、1H NMR、31P NMR等结构表征确证,经HPLC纯度检测。并以胞苷为例探讨了氨基、羟基保护试剂、物质的配比等因素对单体7a收率的影响。

Synthesis of (N,N-Dimethylamino)phosphoryl Chloride Morpholino Nucleoside Monomer

Abstract:(N,N-Dimethylamino)phosphoryl chloride morpholinonucleoside monomers were synthesized through protection of amine group in base, protection of 5'-hydroxyl , transformation of ribose to morpholine ring, N-trityl in morpholine ring, deprotection of hydroxyl group and phosphorylation of hydroxyl group using cytidine and ribothymidine as raw material respectively. The total yield were 20% and 30%, respectively. All intermediates and the target products were characterized by LC-MS, 1H NMR, 31P NMR, and their purity were measured by HPLC. The influence of amino and hydroxyl protective reagents and the ratio of substances on the yield of monomer 7a was also investigated.
