替格瑞洛降解杂质EP-A制备和结构确证

摘 要:以2-[[(3aR,4S,6R,6aS)-6-氨基-2,2-二甲基四氢-3aH-环戊二烯并-1,3-二氧杂环戊-4-基]氧基]-1-乙醇-L-酒石酸盐和2-丙硫基-4,6-二氯-5-氨基嘧啶为起始原料,经缩合、环合、氨解和脱保护4步反应合成得到替格瑞洛降解产生的有关物质EP-A。收率66%,纯度97.06%(HPLC)。其结构经LC-MS、NMR表征确证,为替格瑞洛原料和制剂的质量控制提供参考。

Detection and Preparation Method of Impurity EP-A from the Degradation of Ticagrelor

Abstract:The related substance (EP-A) produced from the degradation of Ticagrelor was obtained by four steps including condensation reaction, cyclization reaction, aminolysis and deprotection using 2-[[(3aR,4S,6R,6aS)-6-Amino-2,2-dimethyltetrahydro-3aH-cyclopentadieno-1,3-dioxolane-4-yl]oxy]-1-ethanol-L-tartrate and 2-propylthio-4,6-dichloro-5-aminopyrimidine as the starting materials. The yield was 66%, and the purity was 97.06%(HPLC). The EP-A was confirmed by LC-MS, 1H NMR. This substance was used as reference for quality control.
