HPLC-RID法测定磷酸奥司他韦干混悬剂中山梨醇的含量

摘 要:磷酸奥司他韦是目前临床上治疗禽流感、甲型H1N1流感病毒和乙型流感病毒最有效的药物之一。采用高效液相色谱-示差折光检测法测定磷酸奥司他韦干混悬剂中关键辅料山梨醇的含量。优选色谱柱Sepax Technologies carbomix Ca-NP 10∶8%(7.8 mm×300 mm,10 μm),流动相为超纯水,流速0.5 mL/min,色谱柱柱温80 ℃,检测器温度55 ℃,进样量20 μL。结果显示:磷酸奥司他韦干混悬剂中除山梨醇之外的其余混合辅料及原料药均不干扰山梨醇的检测;山梨醇在一定浓度范围内呈良好线性关系;重复测试6份样品,相对标准偏差0.53%,回收率99.36%~100.33%,RSD为0.30%。所建立的检测方法适用于磷酸奥司他韦干混悬剂中山梨醇含量的测定。

Determination of Sorbitol in Oseltamivir Phosphate Suspension by HPLC-RID

Abstract:Oseltamivir phosphate is currently one of the most effective drugs for avian influenza, influenza A (H1N1) virus and influenza B virus. The content of sorbitol in oseltamivir phosphate suspension was determined by HPLC-RID on the column Sepax Technologies carbomix Ca-NP 10:8% (7.8 mm×300 mm, 10 μm). The separation conditions included: the mobile phase is water; the flow rate was 0.5 mL/min; the column temperature was 80 ℃; the detector temperature was 55 ℃; and the injection volume was 20 μL. The test results showed that the other mixed excipients and API in oseltamivir phosphate suspension other than sorbitol do not interfere with the detection of sorbitol. Sorbitol showed a good linear relationship. Six samples were tested repeatedly, with a relative standard deviation of 0.53%, the recovery rate is 99.36%~100.33%, RSD is 0.30%. The established method is suitable for the determination of sorbitol in oseltamivir phosphate suspension.
